CJC-1295 and Ipamorelin: Why Researchers Pair Them

CJC-1295 No DAC and ipamorelin turn up together in growth hormone research more than almost any other pair. The reason is simple: they work through two different receptors on the same pituitary cells, so studying them together shows how the two signals combine.

Two receptors, one axis.

Growth hormone release from the pituitary answers to two main signals. One is growth hormone-releasing hormone (GHRH), acting on the GHRH receptor. The other is ghrelin, acting on the growth hormone secretagogue receptor, GHS-R1a.

CJC-1295 No DAC Ipamorelin
Receptor GHRH receptor Ghrelin receptor (GHS-R1a)
Class GHRH analog Growth hormone secretagogue
Length 29 amino acids 5 residues
Also known as Mod GRF 1-29, tetrasubstituted GRF —
Developed by Modified from GHRH (1-29) Novo Nordisk, 1990s

CJC-1295 No DAC stands in for GHRH. Ipamorelin stands in for ghrelin. Because they don’t compete for the same receptor, researchers can look at each signal alone and then at both together, where earlier work reported the two reinforce each other.

What CJC-1295 No DAC is.

It is the 29-residue active fragment of GHRH with four amino acids swapped (D-Ala2, Gln8, Ala15 and Leu27). Those changes protect it from the enzyme DPP-4 and from oxidation, so it lasts longer than unmodified sermorelin.

The name confuses people. The original CJC-1295 from ConjuChem carried a Drug Affinity Complex (DAC), a linker that ties the peptide to albumin in the blood and stretches its half-life to days. No DAC is the same peptide without that linker, so it acts briefly, closer to a natural pulse.

What ipamorelin is.

Ipamorelin is a five-residue peptide from the growth hormone secretagogue family, alongside GHRP-2 and GHRP-6. What made it stand out in its first studies was selectivity: it released growth hormone without the noticeable rises in ACTH and cortisol that earlier GHRPs caused. That is why it became the standard ghrelin-receptor tool in research.

Sermorelin vs ipamorelin.

The comparison comes up often, but the two aren’t alternatives in the way the question suggests. Sermorelin acts on the GHRH receptor, like CJC-1295 No DAC. Ipamorelin acts on the ghrelin receptor. So sermorelin plus ipamorelin is another two-receptor pairing, with sermorelin’s shorter half-life in place of CJC-1295’s more stable profile. For more on the GHRH side, see sermorelin vs tesamorelin.

Further down the axis.

Growth hormone signals onward through IGF-1, mostly made in the liver. Researchers who want to work at that level, independent of the pituitary, use IGF-1 LR3, a long-acting IGF-1 analog that binds poorly to IGF-binding proteins.

Formats and documentation.

CJC-1295 No DAC and ipamorelin each come as lyophilized powder in 5 mg and 10 mg vials. Every lot is third-party HPLC tested, with certificates in our COA library. To prepare them, see how to reconstitute peptides and our bacteriostatic water.

Questions.

Why is ipamorelin paired with CJC-1295 rather than another GHRH analog?

It can be paired with any of them. CJC-1295 No DAC is popular because it is more stable than sermorelin while still acting briefly.

What is the difference between CJC-1295 with and without DAC?

DAC is a linker that binds the peptide to albumin so it lasts for days. Without it, the peptide acts for a much shorter time.

Is ipamorelin a GHRP?

It belongs to the same secretagogue family as GHRP-2 and GHRP-6 and acts on the same receptor, but it was found to be more selective.

Are these approved for human use?

No. Both are sold for laboratory research only.
